Weissman Human CRISPRi Titration Libraries

  • Purpose

    These libraries contain ~5,000 series of systematically attenuated CRISPRi gRNAs targeting ~2,500 genes involved in central biological processes. The activity of gRNAs within each series vary from fully functional to inactive. When paired with CRISPRi, each series enables titration of a targeted gene from normal expression level to completely knocked down.

    The large-scale library (Addgene #136478) contains two series of gRNA per gene with 23–24 gRNA per series.

    The compact library (Addgene #136479) contains two series of gRNA per gene with 1–9 gRNA per series.

  • Vector Backbone

    pCRISPRia-v2 (Plasmid #84832) - does not express Cas9

Library is composed of approximately 5000 sgRNA series targeting approximately 2500 genes. Each series is composed of an original sgRNA, and 22 to 23 variants with mismatches in large-scale libraries, or 2 to 8 variants with mismatches in compact libraries.
