  • Purpose

    This piggyBac barcoding library is a component of the macsGESTALT inducible lineage recording system. The plasmid contains both a static barcode (which is a random 10-mer ‘library’ here) and an evolving barcode (which is edited by CRISPR/Cas9 components delivered by other plasmids).

    A companion plasmid, pUltra-U6-gRNAs1-5 (Addgene #178569, ordered separately), includes a gRNA-tRNA array for macsGESTALT editing.

  • pUltra-U6-gRNAs1-5 (Addgene #178569) and a Cas9 plasmid are NOT included with this item and will have to be ordered separately. The library can be used in conjunction with Lenti-iCas9-neo (Addgene #85400) or any other plasmid(s) or cell lines expressing Cas9.

This library is delivered as suspended DNA in a microcentrifuge tube on blue ice. The tube's contents will not necessarily be frozen. For best results, minimize freeze/thaws.
