Modifications to backbonePlasmid assembled from 3 CYANO-VECTOR modular devices: pCVD019 / CV-S7942NS1-RK2BOM: The device comprises the chromosome integration site NSI for Synechococcus elongatus PCC7942. The device has an origin of replication for E. coli derived from pBR322 and an oriT site for conjugal transfer recognized by the RK2/RP4 mobilization machinery. pCVD065 / CV-aadA_nt: The device confers spectinomycin and streptomycin (SpSm) resistance. About hundred nts of the omega cassette follow the CDS but it does not have a synthetic terminator. Nevertheless this device works well. pCVD015 / CV-ccdB-SwaI: The device comprises the ccdB gene (toxic for commonly used E. coli cell line), which can be taken out with the SwaI endonuclease and swapped for the sequence(s) of interest using adaptors optimized for seamless assembly reaction.
