Description

The MoBiFC toolkit is a collection of Golden Gate/MoClo compatible vectors for assembling single-TDNA bimolecular fluorescence complementation experiments in plants. It includes positive and negative interaction controls for chloroplast BiFC experiments, as well as TurboID modules for proximity labelling experiments.

(A) Outline of a multigenic MoBiFC construct for testing an interaction between two proteins of interest (POIs) by BiFC. The example is shown with chloroplast target peptides (green) and C-terminal YFP fragment fusions (yellow; n, N-ter fragment; c, C-ter fragment). OEP7-TRQ is a reference fluorescent protein for transformed cells. (B) Images displaying BiFC assays between two protein pairs in N. benthamiana mesophyll cells. The HSP21-cYFP HSP21-nYFP interaction allows the reconstitution of the YFP fragments into functional YFP, while the negative control using mCHERRY-nYFP does not. Scale, 10 µm; histogram levels are shown at the bottom of each channel.
