pTF1 requires a reverse primer. This entry [NCBI gibbsq 122487] was created by the journal scanning component of NCBI/GenBank at the National Library of Medicine. This sequence comes from Fig. 1C. Deposited by: Allen Portner High-copy-number plasmid (in bacteria) with increased efficiency of liposome-mediated introduction into cultured cells. [1] Can express foreign sequences in cells producing T7 RNA polymerase, such as that encoded by recombinant vaccinia virus vTF7-3 ATCC VR-2153. [1] Constructed by inserting the 152 bp BamHI-BglII fragment from pAR2529 into the BamHI site of pTZ18R. Polylinker sites were removed and were replaced with the 48 bp EcoRI-KpnI fragment of pBluescript KS+. [1] The order of the major features of the plasmid is: universal primer sequence - XbaI - T7 phi 10 terminator < - XbaI - BamHI/MCS/EcoRI - < T7 phi 10 promoter - reverse primer sequence - pMB1 ori - < ampR - f1 ori.[1]CC A correction to the published sequence has been submitted.
