The following unique restriction sites are found on this vector separated by (bp)(approx): BglI - 100 - PstI - 750 - EcoRI - 600 - XhoI - 300 - SmaI - 250 - HindIII - 1650. [1] Restriction digests of the clone give the following sizes (kb): EcoRI--3.7; PstI--XhoI--2.3, 1.4; BglI--3.7. (ATCC staff) Sequencing has demonstrated that the OL PL region is unchanged from lambda but that one A at position 41 of the 5' untranslated region of the N gene has been deleted. The translational start for the N gene product is missing. [1] The orientation of the kanR segment in pPLa2311 is such that the HindIII site is closer to the ori-containing fragment. [1] The orientation of the PL promoter is anticlockwise with respect to the plasmid ori. [1] This vector has 3 HaeII fragments: one about 2 kb containing PL and beta-lactamase from pBR322, one from pMK20 carrying kanR, and one 370 bp containing the ori segment from pMK20. [1] pPLa2311 in M5219 does not form colonies at 42C, even in the absence of antibiotic selection. Plating efficiency is variable in K12deltaH1 at 42C. [1] E.coli M5219 is E.coli K-12 M72 lac(am) trp(am) rpsL lambda cI857 deltaH1 bio252. deltaH1 removes part of cro and all genes to the right of cro. bio252 removes all genes to the left of cIII.
