- Restriction digests of the clone give the following sizes (kb): EcoRI--6.6, 2.1; HindIII--5.0, 3.9; SstI--9.0. (ATCC staff) Plasmid contains the following restriction sites (approximate kb from nt 1): BamHI--5.91; BglI--5.73, 7.32, 8.53; EcoRI--3.77, 5.88; HindIII--2.25, 5.96; KpnI--5.89; PstI--0.65, 1.72, 5.94, 7.45; SacI--5.88; SmaI--0.93, 4.50, 5.89; XbaI--5.92. (GenBank/EMBL/DDBJ) A series of overlapping deletion segments can be generated by partial digestion with a restriction enzyme not found in the vector, followed by complete digestion with lambda terminase. The order of the major features in the plasmid is: 5' lacZ - Ecl136II/MCS/Sse8387I - 3' lacZ - ren - P - O - ori lambda - cII - cro - kanR - cosR/L - ampR - oriP. [1] Cosmid vector containing two compatible origins of replication, for generating bidirectional deletions of large inserts to facilitate sequencing. Resulting fragments are blunted and circularized by ligation, and transformants are selected for either ampicillin or kanamycin resistance. AmpR transformants can be sequenced in the clockwise direction using the cosL primer 5'-TCATAAATAGCGAAAACC-3'. KanR transformants can be sequenced in the counterclockwise direction using the cosR primer 5'-ACTTTACGGGTCCTTTCCG-3'.
