- These data and their annotation were supplied to GenBank by Will Gilbert under the auspices of the GenBank Currator Program. Revised 16-DEC-1986 by F. Pfeiffer: There is a sequence conflict in that pMAC58 contains a TTG sequence where its parent pBR325 at pos 5264-5265 only contains a TG. It is unclear, if this is a mutation in pMAC58, or if the sequence of pBR325 is incorrect at that position. The plasmid pBR325 is a cloning vector constructed in vitro by the addition of the chloramphenicol resistance gene of an IS1-flanked transposon to pBR322. Sequence analysis suggests that this transposon is related to transposons Tn9 and Tn981 but is longer. No portion of insertion sequence IS1 was found in the genome. This sequence was reconstructed from the genome of pBR322, transposons Tn9 and Tn981, and pBR325 genome fragments. The actual pBR325 and pBR328 genomes may differ from this sequence. In pMAC58, a pBR325-derived vector, a TTG sequence was found where pBR325 has only a TG (pos 5264-65). It is not known if this discrepancy is a mutation or a sequence error in pBR325.
