- Constructed by inserting a 1.2 kb 5'-EcoRI/BamHI-3' fragment containing S. cerevisiae HIS3 into the EcoRI and BamHI sites of pSV2-neo. [1] A 0.3 kb BamHI fragment from BLUR13 was modified by adding an EcoRI site at the SphI site & then cloned into the BamHI site between neoR and HIS3. [1] If linearized with EcoRI and transformed into YAC-containing his3 deletion strains, the plasmid will target integration to Alu sequences and confer a His+ phenotype. The YAC can then be transferred to a eukaryotic cell by spheroplast fusion. [1] YI-type shuttle integrative vector to incorporate a neoR casette into human-derived (Alu-containing) YACs. [1] Restriction digests of the clone give the following sizes (kb): BamHI--6.1, 0.3; EcoRI--6.4; HindIII--3.4, 3.1.
