- Notes
- Plasmid linearized with PstI can be used to introduce cDNA libraries constructed in pcD or pcD2 vectors into Schizosaccharomyces pombe, resulting in the formation, by homologous recombination, of concatamers maintained as episomal DNA. Contains the ars and stb sequences from Schizosaccharomyces pombe and the LEU2 gene from Saccharomyces cerevisiae. The PstI site interrupts the beta-lactamase gene. Shares pBR322 sequences from the PvuII site to the EcoRI site with pcD. Restriction digests of the clone give the following sizes (kb): EcoRI--3.3, 1.3, 1.2, 0.9; HindIII--6.6; PstI--6.6. (ATCC staff) Medium is 1227 LB plus ampicillin. Hosts: E.coli HB101, Schizosaccharomyces pombe, E.coli, Schizosaccharomyces pombe [ATCC 38399].
