Mali Lab Pan-Druggable Cancer Library
(Pooled Library #182133)
-
Purpose
This library provides an all-in-one CRISPR/Cas system composed of 74 elements: 63 sgRNAs targeting cancer-related genes with FDA-approved drug treatments, 3 sgRNAs targeting key medulloblastoma driver genes, 4 sgRNAs targeting the AAVS1 locus, and 4 non-targeting controls.
-
Vector Backbone
The LentiCRISPR v2-GFP (GB, 23 KB) backbone was prepared by replacing the puromycin-resistance domain from LentiCRISPR v2 with an EGFP reporter sequence.

Figure 1: Pan-Druggable Cancer Library design. Left: Library was designed by compiling a list of FDA-Approved cancer drugs, listing the proteins targeted by these drugs, then designing sgRNAs targeting these genes. Right: List of targeted genes, with genes associated as medulloblastoma drivers highlighted in blue. Figure adapted from Hu et al. 2022.
The depositing lab recommends using MAGeCK/MAGeCK-VISPR algorithms (Link opens in a new window) for analysis.
