Purpose
This lentiviral genome-scale CRISPR library targets 18,436 human genes with 185,634 guide RNAs. The library is divided into 2 parts (h1 and h2). The h1 and h2 libraries contain in total 10 gRNAs per gene (5 gRNAs in each half library).
gRNA design for this library is optimized to (1) maximize on-target cleavage effects using the SSC algorithm developed by the Liu Lab ( Xu et al. Genome Research 2015), (2) minimize potential off-target effects, and (3) ensure the fidelity of their sequence with corresponding target loci and avoid regions with possible genomic variations.
