Superti-Furga Lab Human Transporter Knockout Libraries

  • Purpose

    These libraries can be used to test the impact of transmembrane transporters on biological processes. Since the libraries contain significantly fewer sgRNAs than a genome-wide CRISPR library, they are suitable for medium throughput approaches and cell systems where the number of cells cannot easily be scaled up.

    Library #213695 was cloned into a vector that expresses both sgRNAs and the Cas9 protein together with a Blasticidin marker (Plasmid #221432) and can be used for a wide range of cell lines without the need to provide Cas9 from a second vector.

    Libraries #221409 and #221410 are cloned into a vector that expresses sgRNAs together with a geneticin marker, but does not express the Cas9 protein (Plasmid #222694, with modifications). Additionally, these libraries contain unique molecular identifiers (UMIs) that allow tracking of individual clones from pooled sequencing data. Library #221409 contains the same transporter targeting sgRNAs as Library #213695. Library #221410 contains 300 sgRNAs targeting 50 ABC transporter genes and pseudogenes as well as 300 control sgRNAs.

Library Shipping

This library is delivered as suspended DNA in a microcentrifuge tube on blue ice. The tube's contents will not necessarily be frozen. For best results, minimize freeze/thaws.
