Trisomy for human chromosome 21 (Hsa21) results in Down syndrome (DS) which is among the most complex genetic perturbations compatible with survival past term. To advance understanding of DS, the Reeves lab has subcloned 163 individual cDNAs (representing 162 conserved genes from Hsa21) into the pCS2-DEST vector to create an expression-ready clone set.
Individual plasmids from this collection can be utilized for in vitro transcription from the SP6 promoter or transient expression from the CMV promoter in mammalian cells, and the entire clone set is compatible for mass cloning into other vectors.
This kit will be sent as bacterial glycerol stocks in 96-well plate format.

Description
Trisomy for human chromosome 21 (Hsa21) results in Down syndrome (DS) which is among the most complex genetic perturbations compatible with survival past term. While trisomy affects development of every tissue, reduced cognitive ability in DS is among the most limiting features and DS is one of the leading genetic causes of intellectual disability.
Because dosage imbalance in DS is present from conception onward, it is of interest to interrogate the effects of individual and combinatorial gene over-expression at early stages of development, which is difficult in mammalian models. The zebrafish, however, provides a tractable system for a large-scale screen of gene effects on early embryogenesis.
RNAs transcribed in vitro from this cDNA cloneset were injected into zebrafish embryos and individuals were screened for effects on early development (Edie SE et al, manuscript in revision). Several cDNAs consistently resulted in phenotypes accordant with down regulation of the shh pathway, and an additional clone repeatedly resulted in pericardial edema. The Reeves lab foresees the utility of this clone set for use in screens for other phenotypes in various cells and tissues.
