Modifications to backboneNLS-tdMCP was amplified by PCR from phage-UBC-NLS-HA-tdMCP-GFP using NLS-tdMCPF+R primers. To add flanking attL1 and attL2 sites (Hartley et al., 2000; Walhout et al., 2000), PCR fragments were TOPO cloned into pCR8/GW/TOPO (K250020, Invitrogen) to make pME-NLS-tdMCP.
