Growth instructionsTransformation/Growth instructions: The transformation procedure is as following: 1) Place the bacteria on ice for 5-10 min to thaw. 2) Add 50 ng of plasmid and incubate for 10 min on ice. 3) Water bath the bacteria at 42C for 30 sec. 4) Place the bacteria on ice for 2 min. 5) Add S.O.C. media. Incubate 2 hours at 20C. 6) Plate bacteria on the agar plate containing 350 ug/ml of ampicillin. 7) Grow bacteria for 48 hours at 20C. The resulting colonies should be small. 8) Pick the colonies up and grow them for 48 hours in LB medium supplemented with 350ug/ml of ampicillin at 20C.
