Parental vector (pGIPZ containing a non-silencing control hairpin) was cut with XbaI + NotI to remove the CMV promoter and TurboGFP. UCOE / SFFV enhancer promoter complex and mutant ORF with N-terminal TurboGFP were PCR amplified and Gibson cloned into the cut vector.
