Please note- the NdeI site has been disrupted in this plasmid. The NdeI site (CATATA instead of CATATG) will generate His-Ile instead of His-Met upon translation to protein. This region is linker between N-tag and protein of interest, so this should not affect protein function.
nmt1/nmt1*/nmt1** - thiamine repressible promoter wt/weak/very weak, correspondingly. MCS-attR1-CmR-ccdB-attR2 cassette was introduced into pREP-X vectors by XhoI sites.
