This gel depicts the NEB 1kb Extend DNA ladder and a digestion of plasmid 73340 with SpeI-HF and PacI.
We expected the following banding patterns:
SpeI-HF single digest: if correct: 1 band: 10.2kb if wrong: 1 band: 11.6kb
SpeI-HF / PacI double digest: if correct: 2 bands: 8.5 and 1.7kb if wrong: 2 bands: 8.5 and 3.1kb
We see a mixed population of both correct and incorrect plasmid.
