The LOC8R targeting sequence was designed by Benjamin S. Padman during his doctoral research project (Primary supervisor: Georg Ramm)
Insert into an N1 vector by subcloning. The number of Cox8 MTS repeats in LOC8R can be modified as follows:
4x Cox8: Insert at HindIII & AgeI sites. (subclone directly).
3x Cox8: Digest with BamHI and re-ligate. insert at HindIII & AgeI sites.
2x Cox8: Digest with XhoI and re-ligate. insert at HindIII & AgeI sites.
1x Cox8: Digest with HindIII and BamHI. Insert at HindIII & BamHI sites.
