This gel depicts the NEB 1kb ladder and a digestion of the plasmid pHAGE-TO-dCas9-3XmCherry with the following enzymes: AgeI-HF, EcoRI-HF, and BamHI-HF. We expect the following results based on the full sequence:
Digested with AgeI-HF, 2 bands at 10.3 and 2.6 kb.
Digested with EcoRI-HF, 1 band at 12.9 kb.
Digested with EcoRI-HF/BamHI-HF, 2 bands at 8.8 and 4.1 kb.
We see the correct results.
