All restriction sites between EcoRI and HindIII are single-cut except AscI. Multiple gRNA expression cassettes cloned within the MCSs between two AscI sites can be subcloned into the AscI site of pFGC-pcoCas9 binary plasmid (Addgene plasmid#52256) through one-step cloning.
