Modifications to backboneThe sequence ccATGgcagc was added to the Adonis aestivalis cbfd2 cDNA (AdKeto2) by PCR in order to provide first three codons that were missing from this cDNA, and also to introduce an NcoI site at the initiation codon. A 1.12 kb NcoI-XhoI fragment (with the XhoI site provided by the C terminal adaptor used in construction of the cDNA library) was then cloned in-frame in the NcoI and XhoI sites of pBAD/His B.
