the attP region of pDONR221 was amplified with primers H3140 and H3141, incorporating MluI and XhoI sites respectively and cloned into the NS1 vector pAM2314. the attP amplified region contains the upstream and downstream M13F and M13R annealing sites for sequencing inserts
