Addgene Diagnostic Digest
This gel depicts the NEB 1kb ladder and a digestion of the plasmid pFRT/TO/HIS/FLAG/HA-MYEF2 with the following enzymes: BamHI-HF and SpeI-HF.
Expected Bands
BamHI-HF: 7.3 kb
SpeI-HF: 5.2 kb + 2.1 kb; Customer saw large contaminating bands above 5.2 kb in this and the double digest.
BamHI-HF/ SpeI-HF: 5.2 kb, 1.4 kb , and 0.7 kb
The contaminating bands are the correct size expected from digesting the original Gateway Entry Vector.
