Modifications to backboneThe targeting vector was constructed using a 4.0 kilobase (kb) Sac1 fragment as the 5homology arm and a 1.5kb Xba1 fragment as the 3arm. A diptheria toxin (DTA) cassette was used to select against random integration, and positive selection was provided by the neo cassette, flanked by loxP sites for subsequent removal in culture. The Foxd3 targeted allele has the entire coding region deleted as well as 70 bp of the 3 flanking DNA. A Gfp gene with an internal ribosomal entry site (Ires, for the Foxd3Ires-Gfp allele) was inserted as described in Hanna et al.
