Backbone

  • Vector backbone
    pFA6
  • Modifications to backbone
Transformed E. coli cells are resistant to ampicillin. The plasmid contains the URA3 gene from Candida albicans and confers uracil prototrophy of transformed auxotrophic yeasts. In contrast to pAG60 this plasmid contains direct repeats of 500 bp from Aschbya gossypii flanking the insert. Plasmid usefull as template for PCR to create gene deletions. Integrative transformation of the respective PCR fragment and selection of the mutants. Provides the possiblity for a marker rescue by homologous recombination at the direct repeats. Counterselection with floro orotic acid.
