Modifications to backboneTransformed E.coli cells are resistant to ampicillin. The plasmid contains the hph gene from Klebsiella pneumoniae encoding hygromycin B phosphotransferase and confers resistance to the antibiotic hygromycin B of transformed yeasts. Other plasmids conferring hygromycin B resistance: pAG26, pAG32. In contrast to pAG32 this plasmid contains direct repeats of 500 bp from Aschbya gossypii flanking the insert. Plasmid usefull as template for PCR to create gene deletions. Integrative transformation of the respective PCR fragment and selection of the mutants. Provides the possiblity for a marker rescue by homologous recombination at the direct repeats.
