After establishing a cell line (CaCo2) with this plasmid, I noticed that the MCM7-V5 from the plasmid does not bind to the chromatin like the un-tagged endogenous MCM7. The tag may disrupt MCM7’s incorporation into the MCM complex, which is crucial for DNA binding.
