The GFP variant in the plasmid backbone, pCSGFP2, contains the F64L and the S65T mutations resulting in 35 times brighter green emission (as described in Ikawa M et al., Curr. Top. Dev. Biol. 44 (1999), pp. 1–20)
This construct is derived from the (−6 kb/−58)-GFP construct [Addgene plasmid number 30460] by replacing the EcoRI–HindIII fragment (−494 to −58) with the corresponding EcoRI–HindIII fragment (−494 to −207) of the 3Δ207 clone (as described in Zapp D et al., Mol. Cell. Biol. 13 (1993), pp. 6416–6426).
