Addgene Diagnostic Digest
This gel depicts the NEB 1kb ladder and a digestion of the plasmid 7TGP with the following enzymes: EcoRI-HF and NotI-HF.
Expected Bands:
EcoRI-HF 5.7 kb & 2.5 kb
NotI-HF 6.2 kb & 2 kb
While the expected bands are present, some additional bands are also apparent, suggesting the plasmid may have recombined. We recommend screening multiple colonies prior to storage of the plasmids. We also suggest cutting to linearize the vector, gel purifying the large band, and religating to obtain the un-recombined plasmid.
