Fragmid is a modular toolkit that leverages Golden Gate cloning to enable the rapid creation of CRISPR vectors compatible with various delivery approaches, enzymes, and mechanisms. It also includes a set of positive control guides to facilitate the benchmarking of CRISPR technologies.

This kit will be sent as bacterial glycerol stocks in 96-well plate format.

Description

Fragmid consists of a comprehensive array of starting plasmids that can be combined in various configurations:

  • The destination vectors are foundational backbones equipped with essential elements for delivery to target mammalian cells, including lentivirus, AAV, and PiggyBac-based transposon. Fragmid also supports delivery to Drosophila cells.
  • The core of Fragmid's versatility lies in its modular fragments, which are categorized into several types based on their function with CRISPR systems:
    • The guide cassettes contain RNA polymerase III promoters and constant RNA sequences for guide RNA association with Cas proteins. They can include validated targeting guides or cloning sites for custom guides.
    • RNA polymerase II promoter fragments offer various promoter options to drive expression based on cell type and desired expression levels.
    • The N-terminal and C-terminal domains include functional domains for different CRISPR applications, such as nuclear localization signals for knockout, transactivation domains for CRISPR activation (CRISPRa), and repression domains for CRISPR inhibition (CRISPRi).
    • Cas proteins encompass a range of Cas enzymes from different species, including nuclease, nickase, and deactivated versions, as well as engineered variants with altered properties.
    • The selection markers provide antibiotic resistance or visual markers for selection in eukaryotic cells.
  • A basic set of pre-assembled vectors enables users to get started with CRISPR technology. These include vectors to express Cas proteins as well as vectors expressing positive control guides.
  • The online portal is a user-friendly web platform that accompanies Fragmid, allowing users to browse available components, design custom CRISPR vectors by selecting from the modular fragments, and download the assembled vector maps: Fragmid Portal (Link opens in a new window) .
Schematic of Fragmid assembly process from day one to day four and resulting vector architecture.  On day one fragment plasmids plus a destination vector are assembled using Golden Gate (BbsI). On day two the assembled vector containing a Guide (2xBsmBI), Promoter, N-terminus, Cas protein, C’terminus, and 2A-Selection undergoes an exonuclease V cleanup followed by transformation and plating. On day three two colonies per construct are picked, miniprepped and restriction digested for gel validation/whole plasmid sequencing. Below the assembled vector are examples of six vector architectures including pRDA_512 lentivirus, pRDA_722 lentivirus, pRDA_789 CROPseq lentivirus, pRDA_889 AAV, pRDA_575 Piggyback, and pRDA_791 empty plasmid.
