This plasmid contains a mariner transposon with a kanamycin resistance gene and an outward-facing constitutive promoter on an R6k gamma suicide backbone. The backbone encodes for ampicillin resistance and can be propagated in pir+ strains (e.g., One Shot PIR1 competent cells). Kanamycin is used to select pir- cells that have undergone a transposition event.
