Purpose
This retroviral plasmid barcode library is sufficient to label more than 30,000 cells with a barcode collision rate less than 1%. The 3' untranslated region of the EGFP coding sequence contains a 32-bp semi-random barcode before the polyA sequence. Single-cell split barcoding (SISBAR) allows simultaneous capture of the transcriptome and viral barcode sequences to infer cellular identities and lineage information across developmental stages.
