The GFP gene is GFP mut3a from Cormack et al (Gene 173:33-38) and contains engineered ribosome binding site.

DNA inserts can be introduced at any of the polylinker sites upstream of the XbaI site.

Promoter activity can be monitored by measuring GFP synthesis by fluorescence microscopy, fluorimetry or flow cytometry.
