Modifications to backboneThe CMV promoter in pLVX-shRNA2-mcherry was excised with BamHI and AgeI digestion and replaced with the UbC promoter, P2A, the zeomycin resistance gene and T2A in frame with mcherry using NEBuilder HiFi DNA Assembly (NEB). UbC, P2A, and T2A sequences were PCR-amplified from pLV-TetO-hNGN2-eGFP-Puro (Addgene #79823) and the zeocin resistance gene sequence was amplified by PCR from FRT-Em7-Zeo-FRT plasmid (Thermo Fisher).
