SpCas9 endonuclease from the Streptococcus pyogenes Type II CRISPR/Cas system, mutated to improve targeting specificity (Slaymaker et al., 2016) was modified via the introduction of the RuvC nuclease domain-disabling mutation D10A and placed in a pCAG.rBGpA expression plasmid backbone.
