This pBA310 is a modified version of the pDEX777 plasmid (Poon et al. 2012 Open Biology https://doi.org/10.1098/rsob.110037) and was originally published in Nerusheva and Akiyoshi 2016 Open Biology (https://doi.org/10.1098/rsob.150206)

NLS and several restriction enzyme sites were inserted between XbaI and BamHI sites of pDEX777 for overexpressing proteins of your interest with/without GFP and with/without NLS. GFP is in between HindIII and XbaI. NLS from Tbrucei La protein (RGHKRSRE) is located in between AgeI and MfeI.

With NLS:
You can clone your gene of interest using MfeI/EcoRV/NdeI/PacI/AscI/BclI/PmeI/BamHI/AflII

Without NLS:
You can clone your gene of interest using XbaI/ClaI/AgeI and PmlI/MfeI/EcoRV/NdeI/PacI/AscI/BclI/PmeI/BamHI/AflII

Make sure your gene of interest is inserted in frame with GFP and/or NLS. Include start or stop codon as appropriate.
