Depositor Comments
The vector enables chromosomal integration via random transposition, facilitated by transposon Tn5 elements (tnp gene as well as outside end sequences OE-L and OE-R), which define the integron - including P-T7 promoters and a TcR marker. Target genes can be inserted at the I-SceI site. The vector backbone is derived from yCP50-poly (Wang and Stillman 1993, PMID: 8441414), which was obtained from ATCC (87555) via LGC Standards GmbH, (see Domrose et al. 2017, PMID: 29552656), and is equipped on the one hand with the bacterial pMB1 ori, aphII-encoded KmR and mob gene from pBBR1 including its promoter and oriT region, and on the other hand with yeast replication elements CEN4-ARS1 as well as the URA3 marker.
