PTPalpha-HA was amplified from plasmid pXJ41PTPalpha using the 5' primer 5'-CGCCAAGCTTGGCCACCATGGATTCCTGGTTCATTCTTGT-3' and the 3' primer 5'-TGTCAAGCTTCATGCGTAGTCTGCACGTCGTATGGGTACTTGAAGT- TGGCATAATCTGA-3'. The 5' primer contains a HindIII site, a Kozak consensus sequence CCACC and the start codon. The 3' primer contains the coding sequence for the HA epitope YPYDVPDYA, the stop codon and a HindIII site. HindIII-cleaved PCR products were ligated into pTet-Splice. The plasmid contains a CMV promoter under control of regulatory sequences from the tet operon such that, when cotransfected with the transactivator pTet-tTAk (Life Technologies), it expresses protein only in the absence of doxycycline.
