Step1: HindIII-human HHARI (about 700bp, 5’ portion)-EcoRV fragment was made by PCR. Step2: Digestion of pCMV-sport6-HHAR1 with EcoR V + Not I, producing 5.3Kb, 1.2Kb fragments, take back 1.2 Kb fragment. Step3: Digestion of pFlagCMV2 with Hind III + Not I, taking back the vector 5.4 Kb. Step4: Ligation of 3 fragments.
