Puro.let7gsm.Cre was generated by two rounds site-directed mutagenesis of Puro.let7g.Cre with mut1F (5’-GATTCCAGGCTGACCTAGTAGTTTGTACAGTTTGAGGGTCTATGATAC-3’), mut1R (5’-GTATCATAGACCCTCAAACTGTACAAACTACTAGGTCAGCCTGGAATC-3’), mut2F (5’-CAGGAGATAACTGTACAGGCCACTGGGTTGCCAGGAAC-3’), and mut2R (5’-GTTCCTGGCAACCCAGTGGCCTGTACAGTTATCTCCTG-3’).
For lentiviral packaging, you can use pCMV-delta-8.2 (Addgene #8455) and pCMV-VSV-G (Addgene #8454).
