In this collection, each Pro-Code is linked to a nuclear-localizing mCherry protein, which results in the Pro-Codes being concentrated in the nucleus.
These kits will be sent as bacterial glycerol stocks in 96-well plate format.

Description
Kit 1000000177: Pro-Code Vector Kit This collection contains a set of 120 lentiviral vector plasmids. Each plasmid encodes a unique protein barcode (Pro-Code), which can be identified on cells by staining with a cocktail of just 10 antibodies. Each Pro-Code is comprised of a mutant version of the human NGFR gene (dNGFR) fused to a sequence encoding three different linear epitopes. The Pro-Code is expressed from the human EF1alpha promoter. The lentiviral vector also contains the U6 promoter and a unique cloning site for insertion of a CRISPR guide RNA. The Pro-Code vectors can be used to carry out pooled CRISPR screens in which 120 different CRISPR-carrying cell populations can be resolved by staining cells with a 10 antibody cocktail and performing single cell flow cytometry, mass cytometry, or by multiplex imaging. The Pro-Code vectors can also be used for tracking cell clones or cell populations.
Kit 1000000197: Nuclear Pro-Code Vector Kit This collection is a set of 75 lentiviral vector plasmids, each with a gRNA cloning site and a unique barcode (Pro-Code). The Pro-Codes in these plasmids are linked to a nuclear-localizing mCherry protein, resulting in the Pro-Codes concentrating in the nucleus. These plasmids can be used for CRISPR screens and for tracking cell clones or populations. The nuclear Pro-Codes are well suited for in situ imaging detection.
