  • Purpose

    The human CRISPR knockout enAsCas12a sgRNA libraries were designed for assays with limited cell numbers. Both libraries (Humagne Set C and Humagne Set D) target the same 19,755 genes and each gene has one construct with two sgRNAs. Each library also contains 100 non-targeting pairs and 500 one-intergenic-site pairs. The total number of constructs per library is 20,355. Set C contains best-ranked guides 1 and 4. Set D contains best-ranked guides 2 and 3.

These libraries are delivered as suspended DNA in microcentrifuge tubes on blue ice. The tube's contents will not necessarily be frozen. For best results, minimize freeze/thaw cycles.

Sets C and D were designed such that guides 1 and 4 were paired together (set C) and 2 and 3 (set D) rather than the sets described in the publication (1 and 2, set A) and (3 and 4, set B), which are slightly more optimal.
