Depositor Comments

Insert: oocyte cDNA from Melton lambda gt10 library.
Insert size: 1526 bp.
Transcription: linearize with Xba1 and transcribe with SP6.
Sense: plasmid #3 on 5/22/90.
Antisense: plasmid #2 on 5/22/90.

Plasmid History:
Constructed by Randall Moon.
Date of Plasmid and by whom: Randall Moon, 4/9/90.
Species of Sequence Origin: Xenopus.

Notes for Use:
lambda G6 Hind III RF in pGEM4 Blue (3/19/90) isolated and blunted into SP64T.

Note: Hind III cuts 9aa 5' of a met which is used for translation, residing in putative leader sequence. This Hind III site is 13 aa 3' of actual initiation met. Removal of 5' UTR and the amino terminal 13 aa's yield a more active Xwnt, but the assumption is that using the second met, and the half leader sequence yield a wild type wnt after cleavage of the shortened leader.
