This vector allows the multicistronic expression of up to three genes in mammalian cells. The construct contains two 2A peptides coding sequences and three cloning positions flanked by restriction sites:

Position 1 (5' AgeI – 3' EcoRV)
Position 2 (5' XbaI – 3' BamHI)
Position 3 (5' NheI – 3' EcoRI or SaII)

The vector is suitable for transient transfection, but could also be used to produce lentiviral particles for stable expression in mammalian cells. For lentivirus packaging use the additional plasmids psPAX2 (Addgene Plasmid #12260) and pMD2.G (Addgene Plasmid #12259).
