shRNA oligos were first cloned into pRetrosuper-Puro digested with BglII and HindIII.

Next, the AgeI/XhoI fragments from shRNA sequence-containing pRetrosuper-Puro plasmids were cloned into the AgeI/XhoI site of pRetrosuper-GFP.
