Depositor Comments

For the CRX-EnR fusion construct, a 302 bp XmaI fragment (amino acids 7–108) containing the Crx homeobox was fused to the EnR domain. To make the CRX-EnR fusion, the XmaI Crx fragment, an EcoRI-XmaI linker encoding amino acids 2–6 with a NcoI site at the methionine of position 2, an XmaI-ClaI linker, and a ClaI-NotI (partial digestion) 930 bp fragment of the EnR were ligated into a shuttle vector, pSLAX21. The ClaI site inside this construct was designed so that the entire CRX-EnR fusion could be excised as a 1.3 kb ClaI fragment from pSLAX21/CRX-EnR. The ClaI fragment was ligated into pLIA.
