Camargo Lab LARRY Barcode Library Version 1

  • Purpose

    The lineage and RNA recovery (LARRY) library clonally tags cells with DNA barcodes. Reading these barcodes with single cell sequencing allows for reconstruction of genome-wide transcriptional trajectories from differentiating cell populations.

    Each barcode is a 40-mer consisting of 28 random and 11 constant nucleotide bases with the structure:
    NNNNTGNNNNCANNNNACNNNNGANNNNGTNNNNAGNNNN

This library is delivered as suspended DNA in a microcentrifuge tube on blue ice. The tube's contents will not necessarily be frozen. For best results, minimize freeze/thaws.

The pLARRY-EGFP construct contains 3’ LTR, EF1a, EGFP, barcode, pA, 5” LTR. A polyT RT-primer recognizes the 3’ a tail of the barcoded mRNA. When the LARRY LV barcode library is added to HSPCs, barcoded EGFP positive cells result.
