This Human Interferon-Stimulated Gene library was developed in collaboration with the Overbaugh and Paddison labs at Fred Hutch.
This ISG-targeting CRISPR KO library was used in Roesch et al. (2018) to screen for ISGs that inhibit VSV/G-pseudotyped lentiviral particles. The curation of the list of ISGs and the library design and synthesis are described in Ohainle et al. (2018). The same pooled sgRNAs have also been cloned into an HIV-specific CRISPR knockout plasmid, the HIV-CRISPR vector, to create an HIV-specific CRISPR KO library, called the PIKA-HIV (Packageable ISG Knockout Assembly). This CRISPR KO library specific for HIV research is available through the NIH AIDS Reagent Program (Link opens in a new window) (Catalog #13566).
The depositing lab recommends using MAGeCK algorithm (Link opens in a new window) for analysis.
